Ralph, J.L., Evaluation of Supplementation of Oxoid Anaerobe Basal Broth with Oxyrase, A Study by Unipath Ltd, Basingstoke, England, online publication, 15 pp ABSTRACT The aim of this study was to compare Oxoid Anaerobe Basal Broth with and without the addition of Oxyrase (TM) for optimal recovery of a test panel of anaerobic bacteria. Suspensions of 60 anaerobes including 9 Bacteroides spp., 23 Clostridium spp., 3 Eubacterium spp., 8 Fusobacterium spp., 5 Prevotella spp., 4 Peptostreptococcus spp., 2 Peptococcus spp., 2 Veillonella spp., Actinomyces israelii, Bifidobacterium bifidum, Bilophia wadsworthia and Propionibacterium acnes were prepared and inoculated into Oxoid Anaerobe Basal Broth containing 2% v/v Oxyrase. Growth of these suspensions was measured over 72 hours using the Labsystems Bioscreen C microbial growth analyzer and the Sy Lab® Bactrac microbial growth analyzer. The growth curves obtained from the optical density measurements using the Bioscreen particularly with Clostridium spp. and Bacteroides spp. showed that the presence of OxyraseTM improved the recovery and speed of recovery of the strains examined. Impedence values obtained with the Bactrac gave variable results. The addition of OxyraseTM to Oxoid Anaerobe Basal Broth improved growth and recovery of the majority of anaerobic bacteria examined, when incubated in the absence of an anaerobic atmosphere.
