Standard curve Figures 1 and 2 show the typical standard curve obtained with standard vitamin B12 in water plotted against the maximum absorbance. Figure 1 shows all points from 0 ng/ml B12 to 0.2 ng/ml B12: Figure 2 shows the linear relationship between 0 and 0.4 ng/ml B12. Figure 3 shows the typical growth curves obtained from Lactobacillus leichmannii growing in assay medium with varying amounts of B12 standard added. The results clearly demonstrate that, over the range tested, an increase in the concentration of vitamin B12 causes an increase in the growth response of the test organism. This response is linear between 0 ng/ml vitamin B12 and 0.04 ng/ml vitamin B12, thus a calibration curve can be constructed and used to calculate the amount of vitamin B12 in an unknown sample.
